左旋多巴对帕金森病大鼠模型学习记忆能力的影响及其机制 被引量:1

The Effects and Mechanism of Levodopa on Learning and Memory Capacity of Parkinson Disease Rats Induced by 6-OHDA

摘要 目的观察左旋多巴(levodopa,L-dopa)对帕金森病(Parkinson disease,PD)模型大鼠学习记忆能力的影响,并探讨其机制。方法应用6-羟多巴胺(6-hydroxydopamine,6-OHDA)制作PD大鼠模型。将228只造模成功的PD大鼠分为对照组和实验组。对实验组大鼠分别按体质量10、20、30 mg/(kg·d)腹腔注射L-dopa,连续28 d,然后分别于腹腔注射L-dopa后1、3、5、7、14、28 d测定其行为学及血浆同型半胱氨酸(Homocysteine,Hcy)和叶酸水平。同时测定大鼠海马区乙酰胆碱酯酶(acetylcholinesterase,AChE)活力。结果 (1)随L-dopa剂量增大、应用时间延长可明显降低大鼠学习记忆能力(P<0.01),降低大鼠海马区AChE活力(P<0.01),升高血浆Hcy水平,降低叶酸水平(P<0.01)。结论大量使用L-dopa可造成大鼠学习记忆能力明显下降,引起海马区AChE活力下降,其机制可能与升高Hcy有关。 Objective To observe the effects of levodopa on learning and memory capacity of Parkinson disease rats and study their mechanism. Methods The model rats for PD were established through 6-OHDA. The success 228 model rats were randomly divided into 2 groups: the control group (CG), the levodopa group (LG). In addition, the LG group was divided into 3 subgroups with different dosages of levodopa [10 mg/(kg . d), 20 mg/(kg .d) or 30 mg/(kg. d)] administered intraperitoneally to each subgroup of rats for 28 consecutive days. At different time points (1 st day, 3 rd day, 5 th day, 7 th day, 14 th day, 28 th day) after intraperitoneally administration, the rats' behavioral changes and plasma concentration of homocysteine and folic acid were observed. The activity of acetylcholinesterase (ACHE) in the hippocampus were checked. Results (1) The learning and memory capacity of Parkinson disease rats was significantly degraded in coincidence with the dosages of levodopa (P〈0.01). The AChE activity in the hippocampus was also decreased in coincidence with the dosages of levodopa (P〈0. 01). (2) The plasma concentration of homocysteine showed significantly increasing and concentration of folic acid showed significantly decreasing in coincidence with the doses of levodopa (P〈0. 01). Conclusions The large dose usage of levodopa may degrade the learning and memory capacity, and AChE activity in the hippocampus of Parkinson disease rats by increasing the plasma concentration of homocysteine.
