成年大鼠和恒河猴骨髓基质干细胞的体外培养及鉴定 被引量:1

摘要 本实验目的是比较成年大鼠和恒河猴骨髓基质干细胞(MSCs)原代培养的细胞形态和生长状况的差异。分别分离成年大鼠和恒河猴的骨髓,密度梯度离心法分离MSCs,传代培养。在细胞生长过程中显微镜观察两种细胞形态及生长状况的差异,并分别进行成脂及成骨诱导。结果表明,在相同的培养条件下,成年大鼠和恒河猴MSCs原代培养细胞形态比较接近,但细胞原代及传代生长状况有差异。大鼠MSCs原代培养及细胞冻存后复苏的生长均快于猴的MSCs。两种动物的细胞都可以向脂肪和成骨细胞分化。猴的MSCs因其生长缓慢和细胞形态的明显改变,需改良其培养条件,才能用于进一步的研究。 The objective of this experiment was to compare the difference of MSCs cell morphology and growth in adult rats and rhesus. We isolated MSCs of adult rats and rhesus and gave passage. Observed the difference of two kinds of cells in cell morphology and growth under microscope, then induced to fat cells and osteoblast respectively. The results showed at the same culture condition, MSCs of adult rats and rhesus had the same cell shapes, but had difference in primary culture and growth state. The gowth of Rhesus MSCs was slower than that of rat MSCs and cells shapes changed more much, soit need impove the culture condition.
