摘要
[目的]优化魔芋组织培养技术,解决种芋携带病原物与病原物累积,使病害逐年加重,造成损失的问题。筛选出一个适宜的魔芋茎尖试管苗培养基,并优化其激素组成和配比。[方法]以魔芋组培苗丛生芽茎尖为外植体,接种到7个不同配方的培养基上培养,选出适宜的基础培养基;再以之为基础,选择6-BA、NAA、GA3进行3因素3水平正交试验,优化其配比。[结果]MS+2.0mg6-BA+0.2mgNAA+0.2mgGA3+30mg蔗糖+6mg琼脂(pH6.0)是魔芋茎尖试管苗适宜的基础培养基。优化试验表明:2.0mg/L6-BA+0.1mg/LNAA+0.2mg/LGA3的激素组合表现最好。[结论]因子分析认为各激素的最佳组合应为2.0mg/L6-BA+0.1mg/LNAA+0.3mg/LGA。
[Objective ] Optimizing tissue culture technique of Amorphophallus, and resolved the loss problems caused by the seed-tuber carrying pathogen and pathogen accumulated which made disease seriously year by year. Screening a suitable medium of test-tube shoots from stem Apexes of Amorphophallus, and optimizing its hormone composition and proportion. [ Method ] To screen the optimal basic medium, stem apexes from shoot duster were inoculated in seven different mediums. Base on these experiments, the combinations of hormones including 6-BA, NAA and GA3 were optimized by orthogonal experimentation. [ Result] The optimal basic medium of test-tube shoots from stem apexes was MS + 6-BA 2.0 mg/L + NAA 0.2 mg/L + GA3 0.2mg/L + sucrose 30 g/L + agar 6 g/L ( pH 6.0). Then, the results showed that the best combination of hormones was 6-BA 2.0 mg/L + NAA 0.1 mg/L + GA3 0. 2 mg/L by orthogonal experimentation, but the factor analysis showed that the best one was 6-BA 2.0 mg/L + NAA 0.1 mg/L + GA30. 3 mg/L .
