穹窿海马伞损伤对前脑NOS阳性神经元的影响

摘要 本研究用NADPH-d组织化学方法,对成年大鼠一侧穹窿海马个切断后前脑NOS阳性神经元的变化进行了计量学分析。结果表明:损伤1周后,损伤侧内侧隔核和斜角带垂直支的NOS阳性神经元数分别减少到71.79%和85.86%,细胞面积缩小到86.34%和92.71%,细胞灰度值增大到111.86%和120.37%;损伤后2周,损伤侧内侧隔核和斜角带垂直支的NOS阳性神经元数量持续下降,分别减少到63.95%和73.56%,细胞面积缩小到61.78%和89.77%,灰度值增大到116.78%和128.96%;损伤后3、4周,损伤侧两核的NOS阳性神经元的变化和第2周时类似,数值保持在较稳定的水平。提示受损的前脑NOS阳性神经元发生退变。 The fimbria-fornix fiber tract was unilaterlly transected in adult rats by a stereotaxic knife cut. NADPH-diaphorase histochemistry was used to assess quantitatively the change of nitric oxide synthase (NOS)-positive neurons in the forebrain at 1,2, 3,4 weeks after the operation. The number of neuronal loss was 28. 21% of NOS-labelled cells in the medial septal nucleus (MS) and 14. 14 % of NOS-labelled cells in the vertical branch of diagonal band (VDB) on lesion side by 7 days, the average size of the cell in MS was decreased by 13. 66% and 7. 29% of the cells in VDB, but the intensity of the reaction was increased by 11.86% of the cells in MS and 20. 37 % of the cells in VDB. Between 2, 3 and 4 week, the number of NOS-labelled cells was reduced by 36. 05% in MS and 26. 44% in VDB, the average area of NOS-positive cell was decreased by 38. 21 % in MS and 10. 33% in VDB. The intensity of the reaction was decreased by 16. 78% in MS and 28. 96% in VDB. On the contralateral side, the NOSlabelled neurons did not differ from those observed in untreated rats. The results revealed that the axotomized septal NOS-positive neurons presented rapid retrograde neuronal degeneration.
