大鼠部分脑区内一氧化氮合酶与经典神经递质的共存 被引量:19

COEXISTENCE OF NITRIC OXIDE SYNTHASE WITH CLASSICAL NEUROTRANSMITTERS IN SOME REGIONS OF THE RAT BRAIN

摘要 用NADPH-黄递酶(NADPH-d)组织化学结合免疫细胞化学双标记方法,对大鼠部分脑区内一氧化氮合酶(NOS)分别与5-羟色胺(5-HT)、酪氨酸羟化酶(TH)和乙酰胆碱(ACh)等共存神经元进行了观察。结果表明在中缝背核(DR)背外侧区,绝大多数NOS神经元与5-HT共存,但5-HT神经元仍有一半以上未表现出NADPH-d阳性。在脑桥蓝斑(LC)腹内侧部存在大量NOS神经元,其中一小部分与去甲肾上腺素(NA)能神经元(以TH为标记)共存。在中脑黑质(SN)区观察到少量NOS神经元散在地分布干大量的多巴胺(DA)能神经元(以TH为标记)之中,此区还存在大量NADPH-d阳性纤维,但没有观察到NOS与DA共存神经元。在前脑中央隔区(MS)、Broca斜带核水平支(hDB)和垂直支(vDB),92%以上的NOS神经元与ACh共存,这些共存神经元约占所有胆碱能神经元的42%。纹状体内也有不少NOS神经元,但它们不与ACh共存。上述NOS分别与5-HT、NA和ACh等经典神经递质共存的结果,为NO参与多种递质功能的调节提供了形态学基础。 Using NADPH-diaphorase(NADPH-d)histochemistry combined with immunocytochemistry,we investigated the coexistence of nitric oxide synthase(NOS)with serotonin(5-HT),tyrosine hydroxylase(TH)and acetylcholine(ACh),respectively,in some regions of the rat brain;In the dorsloateral subgroup of the dorsal raphe(DR)nucleus,most of the NOS neurons displayed 5-HT immunoreactive(5-HT-IR),while more than half of the serotoninergic neurons were not NADPH-d positive. A large number of NOS neurons presented in the ventromedial subgroup of locus coeruleus(LC), some of them colocalized with noradrenaline(NA)(TH as the maker).In the substantia nigra(SN),NADPH-d+TH double labelled neurons were not observed,although a few NOS neurons containing only NADPH-d activity were visible. Furthermore,there were lots of NADPH-d positive fibers in the SN.In the medial septum(MS),the horizontal and vertical limbs of the diagonal band of Broca (hDB,vDB),more than 92%of NOS neurons displayed ChAT-IR,while such double labelled neurons consisted only about 42%of the cholinergic neurons. A great number of NOS neurons occurred in the striatum,but no NADPH-d+ChAT double labelled neurons were found.The results that NOS coexists with classical neurotransmitters such as 5-HT,NA and ACh in rat brain provide a morphological basis for that NO participates in the modulation of transmitter functions.
