MN9202对兔小肠缺血再灌注损伤时血管内皮细胞功能的保护作用 被引量:7

摘要 目的:探讨二氢吡啶类钙通道阻滞剂MN9202对家兔小肠缺血再灌注引起的血管内皮细胞功能损伤是否具有保护作用.方法:用无创伤动脉夹夹闭兔肠系膜上动脉60min后松夹再灌注,复制小肠缺血再灌注损伤模型.于夹闭45min后给实验组ivMN9202(1μg/kg),手术对照组和休克组给予等量的助溶剂.再灌注1h后处死动物,制备离体血管环,行离体血管功能实验.结果:与手术对照组比较,再灌注组胸主动脉环对低浓度的KCI反应性显著增强,EC50明显降低,对Ach的舒张反应显著减弱.MN9202组与再灌注组比较,胸主动脉环对Ach的舒张反应显著增强(55%±9%υs41%±9,P<O.05,n=6).结论:MN9202有保护血管内皮细胞功能的作用,可能是其对家兔小肠缺血再灌注损伤的保护作用机制之一. Aim: Protective effects of MN9202 on aortic endothelial function in intestinal ischemia reperfusion injury inrabbits was investigated. Methods: Intestinal ischemiareperfusion model was induced by clamping superior mesenteric artery for a 60-min period and then releasing the arterial clamp. MN9202 in a dose of 1 μg/kg was administered in-travenously 45 min after intestinal ischemia. Results: After60 min reperfusion, compared with operative controlgroups, the thoracic aortic vasoconstrictor responses for 20,30 mmol/L KCl in reperfusion group were more significantly augmented, the EC50 values were significantly weaken,and the vasorelaxant action of acetycholine was more significantly impaired. Compared with reperfusion groups,MN9202 could partially attenuate the reperfusion-inducedinhibition of endothelium-dependent relaxation (55 % ± 9%μs 41 %± 9, P<0. 05 ). Conclusion: The beneficial effectsof AN 9202 in intestinal ischemia reperfusion injury in rabbits may be due in part to protect vascular endothelium, andrestore the normal function of vascular constrictionAirelaxation, further resulting in amelioration of the shock state.
