痛泻要方对肠道高敏性大鼠模型肠组织及脊髓后角P物质及血管活性肠肽表达的影响 被引量:12

摘要 目的通过观察肠道高敏性IBS大鼠模型肠组织及脊髓后角P物质(SP)、血管活性肠肽(VIP)免疫反应阳性神经纤维表达的变化,探讨痛泻要方可能的作用机制。方法采用直肠慢性刺激法制作肠道高敏性肠易激综合征模型。将实验动物分为空白对照组、模型对照组、中药低剂量组、中药高剂量组、西药组。观察各组大鼠肠道敏感性指标,用免疫组织化学法对各组大鼠回盲部、结肠及脊髓后角SP、VIP的表达进行半定量分析。结果痛泻要方能提高肠道容量阈值,减少收缩反射次数;降低模型大鼠肠组织及脊髓SP、VIP免疫反应水平,呈一定量-效关系。结论痛泻要方可降低肠道敏感性,其机制可能在于调节肠道初级感觉神经纤维及脊髓后角SP、VIP的分泌,减弱背角神经兴奋性,从而提高内脏痛阈。 Objective To investigate the effect of Tongxieyaofang on the expressions of SP and VIP in rat model of visceral hypersensitivity . Methods The visceral hypersensitivity rats were induced by chronic stress excitation. The rats were divided into five groups : control, model, 15g/kg of Tongxieyaofang, 30g/kg of Tongxieyaofang and Dicetel. The visceral minimum volume threshold and frequency of contraction reflects were observed. The expressions of SP and VIP in ileocecal junction tissue, colon tissue and posterior horn of spinal cord were analyzed semiquantitatively with the immunohistochemistry method. Results Tongxieyaofang could increase the visceral minimum volume threshold and decrease the frequency of contraction reflects. The expressions of SP and VIP in ileocecal junction tissue, colon tissue and posterior horn of spinal cord were lower than those in model group, and it depended on the dose of Tongxieyaofang. Conclusion Adjustment to the secretion of SP and VIP in intestinal tissue and posterior horn of spinal cord may be one of important mechanisms of therapeutic effect of Tongxieyaofang on IBS, which can reduce the excitability of DRG and visceral hypersensitivity.
