支气管哮喘大鼠淋巴液Th1与Th2类细胞因子变化及其与支气管肺泡灌洗液、血浆水平比较 被引量:3

The change of Th1/Th2 cytokines in lymph and comparison of these cytokines in lymph,BALF and plasma in bronchial asthma rat

摘要 为观察哮喘大鼠淋巴液中Th1(IFN-γ)和Th2(IL-4)类细胞因子变化,并比较其与支气管肺泡灌洗液(BALF)、血浆水平间的差异,建立了哮喘大鼠模型。采用ELISA方法检测哮喘组、对照组大鼠激发后0 h、2 h、6 h、10 h、24 h、48 h淋巴液、BALF及血浆IL-4、IFN-γ、IL-12水平。发观对照组和哮喘组大鼠0 h、2 h、6 h、10 h、24 h、48 h各时间点淋巴液中IL-4、IFN-γ、IL-12水平均较BALF、血浆明显升高(P<0.01)。哮喘组大鼠激发后2 h、6 h、10 h、24 h、48 h时间点淋巴液IL-4、IFN-γ、IL-12水平均较0 h明显升高(P<0.05);淋巴液IL-4水平于激发后10 h达高峰,BALF、血浆IL-4水平于激发后24 h达高峰;淋巴液IFN-γ、IL-12水平于激发后6 h达高峰,BALF、血浆IFN-γ、IL-12水平于激发后10 h达高峰。哮喘组大鼠淋巴液、BALF、血浆IL-4水平均较对照组显著升高,而淋巴液IFN-γ、IL-12水平均较对照组明显降低。结果表明,哮喘组和对照组IL-4、IFN-γ、IL-12浓度在淋巴液、BALF、血浆中均由高到低,呈依次递减;且哮喘大鼠淋巴液中IL-4、IFN-γ、IL-12出现高峰均早于BALF、血浆。哮喘大鼠淋巴液存在以Th2亢进为特征的Th1/Th2失衡;淋巴液中细胞因子水平对于反映哮喘炎症程度可能更早和更准确。 To observe the change of Th1(IFN-γ) and Th2(IL-4) cytokines in lymph, and to compare their levels in bronchoalveolar lavage fluid (BALF) and plasma of asthmatic rat, the rat-asthmatic model were established, and the levels of IL-4, IFN g and IL-12 in lymph, BALF and plasma were detected by enzyme linked immunosorbent assay(ELISA) after challenge with ovalbumin in 0, 2, 6, 10, 24, 48 hours in asthmatic and control rats. It was found that the levels of IL-4, IFN-γ and IL-12 in lymph were significantly higher than those in BALF and plasma after challenging in 0, 2, 6, 10, 24, 48 hours in asthmagtic and controls (P〈0.01). In the ashmatic group of rats, the levels of IL-4, IFN-γ and IL-12 in lymph 2, 6, 10, 24, 48 hours after challenge were significantly higher than those in 0 hr (P〈0.05). The level of IL 4 in lymph increased to the maximal level at 10 hour, while that in BALF and plasma increased to the maximal level at 24 h. IFN-γ and 1L-12 in lymph increased to the maximal level at 6 hour, and those in BALF and plasma increased to the maximal level at 10 hour. The level of IL-4 in lymph of asthmatic rats was significantly higher than that in controls, and the levels of IFN g and IL-12 were significantly lower than that in controls. It is evident that the concentration of IL-4, IFN-γ and IL-12 in lymph, BALF and plasma were alike reduced from high to low levels in asthmatic and control rats. The maximal levels of IL-4, IFN-γ and IL 12 in lymph appeared to be early emerged than those in BALF and plasma. From these observations, it suggests that the Th1/Th2 unbalance impressing Th2-hyperfunction exists in lymph of ashmatic rats, and the cytokines level in lymph can reflect the degree of the asmatic inflammation much early and accurately.
