摘要 采用免疫组织化学染色法,检测了61例急性白血病(AL)的p16、bcl-2、bax和fas/apo-1基因表达。结果表明:急性髓系白血病(AML)和急性淋巴细胞白血病(ALL)组的p16、bax和fas/apo-1表达均显著低于对照组(P<0.001). ALL组的 p16、fas/apo-1表达又显著低于AML组(P<0.05)。 AML和 ALL组 bcl-2基因表达均显著高于对照组(P<0.001)。 AML缓解组的 bcl-2基因表达显著低于未缓解组(P<0.01)。提示:AL可同时存在多个凋亡基因缺陷,bcl-2高表达可作为AML患者化疗不敏感的预后指标之一。 By using immunohistochemical staining assay, the expressions of p16、bcl-2、bax and fas/apo-1 genes were investigated in 61 patients with acute leukemia (AL). The results showed that the expressions of p16、bax and fas/apo-1 proteins in acute myeloid leukemia (AML) and acute lymphoblastic leukemia (ALL) groups were significantly lower than those in control (P<0. 01). The expressions of p16 and fas/ apo-1 proteins in ALL were lower than those in AML (P<0. 05). At the same time, bcl-2 expression in AML and ALL was strikingly higher than those in control (P<0. 001). In AML, bcl-2 expression in complete remission group was obviously lower than that in resistant disease group (P<0. 01). The results of above genes expressions indicate that some genes have their apoptotic defect in AL. High expression of bcl-2 gene may be a predictor of resistance to chemotherapy for AML.
