摘要
目的探讨腹主动脉缩窄法模拟压力超负荷大鼠模型对肾细胞凋亡影响及替米沙坦干预作用。方法将36只大鼠随机分成3个组:假手术(SG)组,腹主动脉缩窄(operation group,OG)组,替米沙坦+腹主动脉缩窄(TM+OG)组,每组12只。采用腹主动脉缩窄法复制压力超负荷大鼠模型,TUNEL法测肾脏原位细胞凋亡,免疫组化法观察细胞色素C和bcl-2的表达。结果SG组可见少数凋亡细胞;OG组有较多凋亡细胞,且多集中于肾近曲小管部位;TM+OG组也可见一些凋亡细胞,但明显少于SG。SG组细胞色素C呈弱阳性表达;OG组呈强阳性表达,较集中于肾近曲小管;TM+OG组呈阳性表达,也集中于肾近曲小管。SG组少有bcl-2蛋白表达;OG组部分细胞呈阳性表达;TM+OG组bcl-2蛋白呈强阳性表达,明显强于OG组,集中分布于肾近曲小管部位。结论替米沙坦对腹主动脉缩窄法复制压力超负荷大鼠所致肾组织细胞凋亡具有一定的抑制作用;替米沙坦具有稳定线粒体膜、保护线粒体功能和上调bcl-2蛋白表达的作用,而替米沙坦抗压力超负荷所致肾组织细胞凋亡可能与上述作用有关。
Objective To study the effect of the pressure-overloaded model,produced by abodominal aorta constriction,on apoptosis of kidney tissure,as well as intervention of telmisartan. Methods Thirty-six rats were divided into three groups randomly,12 in each group:SG,OG and TM+OG. Abodominal aorta constriction was adopted to build pressure-overloaded rat model. The de gree of apoptosis was assessed through the method of TUNEL. The expression of cytochrome C and bcl-2 was observed through immunohistochemistry. Results Few cells were detected in SG. There were more cells in OG which mostly gathered around renal proximal tubules. Cells could also be found in TM+OG,but for less than that in SG. Cytochrome C assumed weak positive expression in SG, strongly positive in OG and positive in TM+ OG. Both in OG and TM+OG, cytochrome C gathered around renal proximal tubules. There were little expressions of bel-2 protein in SG. In OG,only part of bcl -2 protein assumed positive expression. TM +OG took on strong positive expression,much stronger than OG and gathered around renal proximal tubule. Conclusion Telmisartan could inhibit the renal cell apoptosis induced by pressure-overlodes produced by abdominal aorta constietion. Meanwhile,it could stabilize and up regulate the expression of bcl-2 protein. The above functions of telmisartan may justify its protection against apoptosis of kodney tissure caused by pressure overloads.
