盐生卤虫BADH基因的克隆及序列分析 被引量:1

摘要 实验以盐生动物卤虫(Artemia)为材料,采用酚-氯仿法提取总DNA,以已报道的BADH基因的保守序列设计引物,通过PCR技术,从卤虫的基因组DNA中分离得到了1个BADH基因片段,经测序,得到一段868 bp的核苷酸序列,输入Gen-Bank进行BLAST同源性比较,发现所得片段与植物中的籽粒苋的BADH4基因中的一段序列有96%的同源性,此项研究为更深入地研究抗盐动物的BADH基因奠定了良好的基础. The total DNA of Artemia was extracted,and one partial BADH were cloned from Artemia through PCR technique by specific primers designed on the basis of the reported BADH gene,which is from 900 bp to 1000 bp.This fragment was inserted into pGM-T easy vector,then introduced into the competence cells of DH5α,and screened of doubtful strains through blue-white spot screening.The combination plasmid was detected by PCR and enzyme digestion,and the suspected strains were sent to sequence.BLAST search revealed that the fragment is about 96%homologous to that of the BADH gene of plant Amaranthus hypochondriacus.This research will help study BADH gene of the salt-resistant animals.
