摘要
目的观察组胺H3受体拮抗剂和激动剂在吗啡依赖大鼠复吸中的作用及相关机制。方法将实验大鼠分为正常对照组、吗啡复吸组、组胺H3受体拮抗剂+吗啡复吸组、组胺H3受体激动剂+吗啡复吸组,通过条件位置偏爱实验建立吗啡复吸大鼠模型,然后分离大鼠的伏核和海马,用放射免疫法检测cAMP的含量。结果组胺H3受体拮抗剂组伏核及海马cAMP含量低于吗啡复吸组(P<0.05),而与正常对照组比较无显著性差异(P>0.05);组胺H3受体激动剂组伏核和海马cAMP含量高于正常对照组(P<0.05),而与吗啡复吸组相比无显著性差异(P>0.05)。结论组胺H3受体拮抗剂可抑制吗啡复吸,其机制可能与其降低大鼠伏核及海马cAMP的水平有关。
Objective To observe the role and related mechanism of histamine H3 receptor on cAMP content in morphine relapse rats.Methods Experimental rats were randomly divided into control group, morphine relapse group, histamine H3 receptor antagonist + morphine relapse group, histamine H3 receptor agonist + morphine relapse group. The rat model of morphine relapse was established by conditioned place preference test. The rat nucleus aecumbens and hippecampus was separated from rats of different group and used to detect the content of eAMP by radioimmunoassay.Results The cAMP content of the nueleus accumbens and hippocampus of histamine H3 receptor antagonist group were lower than morphine relapse group (P〈0.05),and there was no difference comparing with control group(P〉0.05).The cAMP content of H3 receptor agonist group were higher than control group (P〈0.05),while there was no difference comparing with the morphine relapse group (P〉0.05).Conclusions Histamine H3 receptor antagonists ean inhibit morphine relapse and the mechanism may be related to reducing the cAMP content of morphine relapse rat.
