摘要
目的:研究蕲艾提取液对免疫性肝纤维化大鼠Ⅰ、Ⅲ型胶原及TIMP-1表达的影响。方法:W istar大鼠60只随机分为正常对照组、模型组和高、中、低剂量蕲艾提取液治疗组,采用异种血清腹腔内注射构建肝纤维化大鼠模型,肝组织切片HE染色后,光镜下观察大鼠肝组织纤维化程度;SABC型免疫组化染色法观察肝组织Ⅰ、Ⅲ型胶原及TIMP-1含量变化。结果:蕲艾提取液各治疗组与模型组相比,肝纤维化程度显著减轻;肝纤维化模型组及蕲艾提取液治疗组Ⅰ、Ⅲ型胶原含量及TIMP-1水平显著高于正常对照组;蕲艾提取液治疗组Ⅰ、Ⅲ型胶原及TIMP-1水平又显著低于肝纤维化模型组。结论:蕲艾提取液能够显著降低免疫性肝纤维化大鼠Ⅰ、Ⅲ型胶原及TIMP-1表达。
Objective： To study the effect of Artemisia Argyi extracting solution on expression of collagen Ⅰ and Ⅲ, TIMP-1 in immunological hepatic fibrosis rats. Methods： Rat＇s liver fibrotic models were established by intraperitoneal injection with heteroserum. Sixty experimental rats were randomly divided into normal control group, model control group, Artemisia Argyi extracting solution high, middle and low-dose group. After the liver tissue was stained with hematoxylin and eosin （ HE）, the collagen proliferation of the liver tissue were observed under the light microscope; the contain of collagen Ⅰ and Ⅲ, TIMP-1 were observed by immunalhistochemistry, the results of which were analysed by image system. Results： Compared with the model group, the degree of hepatic fibrosis in rats of therapeutic groups reduced significantly ; the contains of collagen Ⅰ and Ⅲ, TIMP-1 in the model group of hepatic fibrosis increased more significantly than that of the normal group, and the contains of collgaen Ⅰ and Ⅲ, TIMP-1 in therapeutic group decreased significantly than that of the model group. Conclusions： Artemisia Argyi extracting solution can reduced the expression of collagen Ⅰ and Ⅲ, TIMP-1 immunological hepatic fibrosis rats.
