摘要 目的探讨3种透明质酸合成酶基因(HAS)在甲状腺相关眼病(TAO)患者和正常人眼眶结缔组织中的表达。方法收集14例TAO患者(TAO组)和8例正常人(对照组)眼眶结缔组织标本,采用逆转录-聚合酶链式反应(RT-PCR)分别检测HAS1、HAS2和HAS3的mRNA表达水平。结果3种HAS基因mRNA在TAO患者和正常人眼眶结缔组织中的检出率相当,差异无显著性意义(P>0.05)。其中,HAS3在全部组织块中检出,HAS1在TAO组为64.3%,对照组为50%;HAS2在TAO组为21.4%,对照组为37.5%;TAO患者眼眶结缔组织HAS3基因表达量为正常人的4.5倍(P<0.01)。结论HAS1和HAS3基因mRNA在TAO患者眼眶结缔组织的大量表达,可能是TAO患者眼眶局部组织中透明质酸增加的分子基础。 Objective To measure the expression of hyaluronan synthase ( HAS) -1 ,-2, and -3 genes in the orbital connective tissues from the patients of thyroid-associated ophthalmopathy ( TAO ) and normal subjects. Methods The expression of HAS-1,-2, and -3 in the orbital tissues from 14 TAO patients and 8 normal subjects was measured by real-time PCR. Resutls HAS3 mRNA was detected by RT-PCR in all tissue blocks from TAO patients and normal subjects. HAS1 mRNA was detected in 9 blocks from the 14 patients,and 4 from the 8 normal subjects. HAS2 mRNA was detected in 2 blocks from the patients,and 3 from the controls. But the differences in the positive rates were not significant. The quantity of HAS3 mRNA copies in patients was 4. 5 times of the normal subjects, respectively( P 〈 0. 01 ). Conclusions Our observations suggest that the increased expression of the HAS genes in orbital connective tissue may be the molecular basis at least in part for the increased accumulation of hyaluronan in orbital connective tissue in TAO.
