摘要
目的探讨雷公藤对哮喘的治疗作用及对痰液中嗜酸细胞(EOS)IL-4和IL-5mRNA表达的影响。方法12例哮喘患者的诱导痰标本,与雷公藤内酯醇(10-7mol/L)共同培养48小时设为雷公藤组(12例),单独培养48小时设为空白组(12例),健康自愿者痰液为正常对照组。采用斑点分子杂交检测EOS IL-4、IL-5mRNA表达。结果哮喘患者痰液中EOS浸润密度与正常对照组比较,差异有显著性意义(P<0.01);雷公藤组与空白组EOS IL-4、IL-5mRNA表达水平比较,差异有显著性意义(P<0.01),空白组与正常对照组EOS IL-4、IL-5mRNA表达水平比较,差异有显著性意义(P<0.01);雷公藤组与正常对照组EOS IL-4、IL-5mRNA表达水平比较,差异无显著性意义(P>0.01)。结论IL-4、IL-5mRNA表达增加可能是导致哮喘嗜酸气道炎症的原因,雷公藤内酯醇能抑制IL-4、IL-5mRNA表达,可能为哮喘抗炎治疗的机制之一。
Objective To explore the role of IL-4, IL-5mRNA in Eosinophil inflammation of asthma airway and effects of triptolide. Methods The expression of IL-4, IL-5 mRNA in EOS cells of the sputum of asthma was measured by dot hybridization after culture of 48 hours in the presence of triptolide (n = 12) and sputum alone as blank group(n = 12). Results The infiltration density of EOS in the sputum of asthma patients was significant different from normal control group. There were significant differences of the expression of IL-4, IL-5 mRNA in EOS between the study group and the blank group or between control group and blank group( P 〈 0.01 ). However, there was no difference of expression of IL-4, IL-5 mRNA in EOS between the study group and in the normal control group ( P 〉 0.01 ). Conclusions The increase of expression IL-4, IL-5 mRNA may play a role in Eosinophil inflammation in airway of asthma. With inhibition of the expression of IL-4, IL-5 mRNA, triptolide might be of potential value in anti-inflammation for asthma.
