摘要
目的为给矮牵牛试管苗工厂化生产提供依据,研究了影响矮牵牛试管苗生长主要因素,包括激素浓度配比、蔗糖和琼脂浓度.方法以无菌种子获得的试管苗为试材,筛选矮牵牛快速繁殖最适培养基.结果IBA浓度为0.3mg/L、6-BA浓度为0.6mg/L时,幼苗生长健壮,长势强,茎干粗壮,叶色绿且叶片厚f蔗糖浓度为20~50g/L时,矮牵牛幼苗生长情况较好;琼脂浓度为6g/L时矮牵牛幼苗生长较快,长势好.结论矮牵牛是一种比较容易进行组织培养的植物,在MS+IBA0.3mg/L+6-BA0.6mg/L+蔗糖20~50g/L+琼脂6g/L,PH6.5条件下增殖系数可达5.00.
Objective To provide evidence for industrialized production of Petunia hybrida Vilm. , the factors affecting in vitro plantlet buds propagation were studied, including concentration of hormones, sucrose and agar. Methods The optimum medium of shoot induction was selected using seedlings of aseptic seeds of Petunia. Results The plantlets had better growth, thickset shoot and dark green and thick leaf when the medium was supplemented with IBA (0.3mg/L) and 6-BA (0.6 mg/L). The seedling grew well when the concentration of sucrose and agar were 20~50 g/L and 6 g/L respectively. Conclusion The Petunia was cultured easily by tissue culture, and the multiplication coefficient was 5.00 in the MS + IBA 0. 3 mg/L q- 6-BA 0.6 mg/L + sucrose 20~50g/L + agar 6 g/L.
