摘要
用两步PCR法,成功地从石蜡包埋组织DNA中扩增了P16INK4exon2片段;并用单链构象多态(SSCP)分析PCR产物的差异.在32例肝癌原发灶中,发现P16INK4exon2缺失突变6例、SSCP可检出突变1例;在48例肝癌转移灶中,发现P16INK4exon2缺失突变11例、SSCP可检出突变2例.
The P16INK4exon2 has been successfully amplifed by two-step-PCR from tissues embeded in parraffin, and the differences of the PCR products were analyzed by SSCP. Six deletions detected by PCR and one SSCP-detectable mutation were found in 32 idiopathy liver cancers while eleven deletions detected by PCR and two SSCP-detectable mutations in 48 liver cancers.
