摘要
目的:建立人体血浆中同时测定咪达唑仑和阿曲库铵HPLC测定法。方法:采用HPLC法,色谱柱为HypersilC18柱(250mm×4.6mm,5μm),流动相为甲醇-水(60∶40)(含0.1%二乙胺,0.05%磷酸溶液),流速为1.0mL·min-1,柱温为室温,检测波长225nm。结果:咪达唑仑在0.25~4μg·mL-1内呈良好的线性关系(r=0.9997),阿曲库铵在6.00~96.00μg·mL-1内呈良好的线性关系(r=0.9998),咪达唑仑和阿曲库铵最低检测浓度分别为50,1.5μg·mL-1,方法回收率在96.8%~100.0%,萃取回收率在87.7%~93.0%。日内、日间RSD(n=3)均小于5.9%。结论:本方法简便、准确,可靠,可用于人体血浆中同时测定咪达唑仑和阿曲库铵的浓度。
Objective:To develop an HPLC method for the determination of midazolam and atracurium in human plasma.Method:The analytical column was packed with Hypersil C18 (250 mm×46 mm,5 μm)with the mobile phase of methanol-water(60∶40),(containing 01%diethyl amine,005%phosphoric acid)at the flow rate of 1 mL·min^-1,and detection wavelength was set at 225 nm.Results:The linear ranges were 025-4 μg·mL^-1 for midazolam in human plasma and 600-9600 μg·mL^-1 for atracurium.The correlation coefficients were 09997 and 09998The midazolam low limit of detection was 50 ng·mL^-1.The atracurium low limit of detection was 15 μg·mL^-1.The method recoveries were 968%-1000%.The extraction recoveries were 877%-930%.Within-day and between-day RSD(n=3)were less than 59%.Conclusion:The method is simple,reliable and sensitive for determination of midazolam and atracurium in human plasma simultaneously.
