摘要
目的探讨大鼠骨髓间充质干细胞(BMSCs)的分离纯化和定向成脂诱导。方法采用密度梯度离心法分离纯化骨髓间充质干细胞,免疫组织细胞化学法检测BMSCs表面抗原。脂肪诱导剂诱导BMSCs分化为脂肪细胞,利用油红O染色进行检测。结果免疫组织细胞化学检测结果显示,BMSCs表达CD29、CD44、CD90、CD106,不表达CD34、CD45。BMSCs经脂肪诱导剂诱导后,油红O染色阳性。随着诱导时间的延长,油红O阳性细胞比例增加。结论体外培养的BMSCs在一定诱导条件下能够向脂肪细胞分化,并且随着诱导时间的延长,脂肪细胞比例不断增加。
Objective To study isolation and ex vivo expansion of bone marrow mesenchynal stem cells (BMSCs) and differentiation of BMSCs into adipocytes. Methods BMSCs were isolated by density gradient centrifugation. Cell surface antigen was confirmed by immunohistochemistry and cytochemistry. Adipogenic differentiation of BMSCs was analyzed by oil Red O staining for lipid accumulations. Resuits Immunohistochemistry and cytochemistry showed that BMSCs expressed CD29,CD44,CDg0 and CD106, but not CD34 and CD45. Oil O staining thowed that BMSCs treated with adipogenic conditions contained lipid droplets and the deoplets increased with time. Conclusion BMSCs under adipogenic conditions can be differentiated into adipocytes and show increased lipid accumulation expression.
