摘要
目的:研究胃腺癌组织和细胞中是否存在分泌血管活性肠肽(vasoactive intestinal peptide,VIP)的肿瘤细胞,以探讨VIP在胃腺癌发生及发展中所起的作用。方法:通过免疫组织化学和RT-PCR检测正常胃组织、胃腺癌组织和培养的胃腺癌细胞中VIP蛋白和mRNA表达情况。结果:胃腺癌组织出现VIP免疫反应阳性细胞,相对正常的胃组织中均未见VIP阳性细胞,胃腺癌组织中无阳性神经纤维,而相对正常的胃组织中均有阳性神经纤维存在,有显著性差异(P<0.01)。培养的胃腺癌细胞中均有VIP免疫阳性细胞染色;胃癌组织和培养的胃腺癌细胞中均有VIPmRNA表达。结论:胃腺癌组织和细胞中含有分泌VIP的肿瘤细胞,VIP在胃腺癌的发生和发展中起着至关重要的作用。
Objective: To study the expression of vasoactive intestinal peptide (VIP) in gastric adenocarcinoma, and evaluate the role of VIP in tumor genesis and progress of gastric adenocarcinoma. Methods: The expression of VIP protein and mRNA in the normal gastric tissue, gastric adenocarcinoma and cells was detected by immunohistochemical staining and RT-PCR. Results: The tissues of gastric adenocarcinoma had the positive cells stained by anti-VIP antibody, but the normal gastric tissues had no such positive cells. The normal gastric tissues had the positive nervous fibers, but the tissues of gastric adenocarcinoma had no such positive nervous fibers. The difference was statistically significant (P〈0.01). There were VIP immunopositive cells in the gastric adenocarcinoma cells. Gastric tumor tissues and gastric adenocarcinoma cells could express VIP mRNA. Conclusion: The gastric adenocarcinoma has tumor cells secreting VIP, which indicates that VIP plays an important role in tumor genesis and progress of gastric adenecarcinoma.
