摘要
目的:建立经济实用的胎儿宫内窘迫动物模型。方法:利用35只20天胎龄的胎鼠,钳夹进入每个胎盘的分支血管,共15分钟,然后松开血管夹,恢复血流,对胎鼠断头处死后进行显微镜检查和测定脑细胞内外钙、钠、钾含量。结果:再灌注15分钟后处死而进行显微镜检查的全部25只胎鼠脑组织均显示结构紊乱,间质和星形细胞水肿,以及神经细胞变性,6/25只(24%)出现神经细胞坏死,7/25只(28%)脑出血。另10只胎鼠在再灌注30分钟后处死进行Fura-2AM法检测,结果显示脑细胞内出现钙超载。结论:此模型可以较好地用于胎儿宫内窘迫脑损害的进一步研究。
To establish an economical and practical animal model for distressed fetus in utero. Methods:Thirty - five fetal rats at twenty days of gestational age were employed for the animal model of distresed fetus in utero. Blood vessels from the branhing point, turning into individual placenta in one hom, were clamped for fifteen minutes. Circulation was then restored for fifteen and thirty minutes respectivelly by removal of the clamps before fetal rats were sacrificed and examined microscopically or biochemicallly. Results: Microscopical examination exhibited structural irregularity, interstitial edema, asteroid cellular edema of cerebral tissue, and neuronal degeneration in cerebral tissue of twenty - five fetal rats(fifteen minutes of reperfusion).And six out of twenty - five fetal rats were found with neuronal necrosis and seven with cerebral hemorrhage. Examination with FUra - 2 revealed that there were intracellular calcium overloads in cerebral cells. Conclusion: This model could be employed in multitudinous experiments conceming cerebral in injury in distessed fetus in utero.
