摘要 采用2,4二硝基氟苯作为衍生化试剂,16种常见人体生理性氨基酸为分析对象,区带毛细管电泳为分离模式,建立一种新的氨基酸衍生化方法。系统研究了反应时间、反应温度和pH值对衍生化反应的影响。同时观察了4℃和25℃室温下衍生化产物的稳定性情况,结果发现,无论是在4℃冰箱,还是在25℃室温下,氨基酸衍生化产物保存5天,紫外吸收值无明显改变。方法的性能指标测定表明,该法的线性在10~700μmo1/L范围内良好(γ在0.964~0.995之间)。最低检测限(S/N=2)为2.5~7.9μmol/L。测得平均回收率在86.3~107.4%之间。该法具有衍生化反应操作简便、灵敏度高、稳定性好、线性及回收率佳等特点,不仅可用于氨基酸的衍生化反应,而且同样适合于其它含有氨基的生物活性物质的衍生化反应。 novel derivative method for the determination of amino acids was developed by the use of 2,4 dinitrofluorobenzen.16 kinds of amino acids were analyzed with high performance capillary electrophoresis.The conditions of derivatization were studied to find an optimal procedure,which included the reaction time,reaction temperature and pH value.The observation of stability shows the products of derivatization of amino acids stay steady for five days at 4℃ or 25℃.The linearity of this assay was 10~700μmol/L,and the relative coefficients were from 0.964 to 0.995.The minimal detection limit(S/N=2) was 2.5~7.9μmol/L.The average recoveries were from 86.3 to 107.4%.The method has many advantages such as rapidity,accuracy,simplicity,sensitivity and stability,which suits to the derivatization of amino acids and other substances which have amino group.
