摘要
目的:观察黄瓜香含药血清在体外对K562白血病细胞增殖及凋亡的影响。方法:采用血清药理学方法制备黄瓜香含药血清,以不同浓度的含药血清处理体外培养的K562白血病细胞,采用MTT比色法观察黄瓜香含药血清对K562细胞增殖的影响,采用wright一Giemsa染色观察肿瘤细胞形态学变化.采用caspase-3活性检测试剂盒检测K562细胞内caspase-3的酶活力水平。结果:不同浓度黄瓜香含药血清对K562细胞增殖具有抑制作用,并呈剂童依赖关系。高剂量黄瓜香含药血清对K562细胞形态学有明显的影响.在黄瓜香含药血清处理K562细胞24 h后,caspase-3活性均显著增加,随黄瓜香含药血清浓度的升高呈现较好的剂量依赖性。结论:黄瓜香含药血清具有抑制K562白血病细胞增殖及诱导凋亡的作用,其作用强度与时间一浓度呈正相关,其作用机理可能与其直接细胞毒作用及提高caspase-3酶的活性有关。
Objective： To study the effect of the herb serum containing VDG on the proliferation and apoptosis of K562 leukemia ceils in vitro. Methods： Prepares the herb serume containing VDG with serologic pharmacological method and treated the K562 leukemia cells with contained herb serum of VDG in different concentration. The effects of the herb serumon the proliferation of K562 cells are observed with MTT assay and the effects of the herb serum on the morphology of K562 cells are observed with Wright -- Giemsa dye. Caspase-3 enzyme activity was measured by caspase-3 enzyme activity detection kit. Results：The herb serum containing VDG in different concentration could inhibit the proliferation of K562 leukemia ceils directly in a dose-dependent manner, caspase-3 activity were significantly increased after treatment with the drug-serum of VDG at 24h in a better dose-dependent manner.Conclusion： the herb serum containing VDG significantly inhibits the proliferation and induces apopotosis in K562 cells. Mechanisms may be involved in accumulation of cytotoxie action and increased caspase-3 activation.
