二色胡枝子遗传转化中抗生素浓度优化研究 被引量:2

摘要 探讨了卡那霉素、G418和头孢霉素3种抗生素对二色胡枝子不同培养阶段外植体生长、分化和生根的影响及对农杆菌的抑菌效果,确立了由农杆菌介导的二色胡枝子遗传转化研究中抗生素转化体的筛选浓度和头孢霉素对菌株的抑菌浓度。结果表明:在子叶节转化筛选阶段,卡那霉素和G418的适宜浓度分别为20 mg/L和7.5 mg/L;在抗性芽增殖培养阶段,卡那霉素和G418浓度分别为150 mg/L和15.0 mg/L;在生根阶段,卡那霉素和G418浓度分别为20 mg/L和5.0mg/L。而头孢霉素在各阶段适宜浓度为0~300 mg/L;头孢霉素对农杆菌LBA4404和C58抑制浓度为200~300 mg/L时效果理想。 In this study, we carried out a research about the effects of three antibiotics of kanamycin, G418 and cephalothin on growing, differentiation and rooting of tissue culture seedling of Lespedeza bicolor ,and we also determined inhibitive effect of cephalothin on the growth of Agrobacterium. After data analysing,we established the optimal antibiotics concentration at different stages for transgenic explant selection during genetic transformation of Lespedeza bicolor conducted by agrobacterium. The results were as follows: In the stage of cotyledon node, the optimal concentration of kanamycin and G418 were 20 mg/L and 7.5 mg/L, respectively. In the stage of buds muhiplication, the concentrations of kanamycin and G418 were 150 mg/L and 15.0 mg/ L. During the rooting stage, the proper concentration of kanamycin and G418 were 20 mg/L and 5.0 mg/L correspondingly. The optimal concentration of cephalothin during each stage was 0-300 mg/L. And when the concentration of cephalothin reached 200~300 mg/L, it could inhibit agrobacterium LBA4404 and C58 growing considerably.
