摘要
本试验利用脂多糖(LPS)诱发试验性大鼠乳腺炎,探讨中药荔蒲煎液对试验性乳腺炎的防治作用。24只SD孕鼠随机分成阴性对照组(NC)、阳性对照组(PC)、药物预防组(Pre)和治疗组(Tre)。Pre组大鼠灌服荔蒲煎液,持续4d。随后,PC组和Pre组大鼠乳房注入LPS,NC组乳管注入等量生理盐水;Tre组大鼠乳管注入LPS24h后灌服荔蒲煎液,连续灌胃4d。对所有大鼠乳腺组织切片进行组织病理学观察、测定胸腺指数和脾脏指数,并进行白细胞计数以及碱性磷酸酶(ALP)和谷胱甘肽过氧化物酶(GSH-Px)的活性检测。结果表明,Pre和Tre组胸腺、脾脏指数以及GSH-Px活性均明显高于PC组(P<0.05),而白细胞数与ALP活性则低于PC组,Pre和Tre组之间无明显差异。病理组织学检查显示荔蒲煎液能减轻LPS引起的炎症反应。上述结果提示荔蒲煎液有利于维持机体的氧化-还原平衡,对LPS诱发的试验性大鼠乳腺炎有较好的防治效果。
This study was conducted to exam the preventive and therapeutic values of Salvia plebeian-Dandelion decoction(SPD) on rat mastitis induced by Lipopolysaccharide(LPS). Twenty-four SD rats were randomly divided into 4 groups, they were negative control group (NC), positive control group (PC), prevention group (Pre) and treatment group (Tre) respectively. Pre was given SPD, NC and PC were given equivalent saline intragastrically for 4 days, and then the rats in Pre and PC were given LPS by injecting into mammary gland to establish mastitis model; NC was injected with equivalent amount of saline. The rats in Tre group were given LPS by injecting into mammary gland, after 24h, the rats were fed with SPD for 4 consecutive days. At the end of experiment, histopathological studies of mammary gland were conducted, thymus and spleen index were calculated, red cells and leucocytes were counted, and the activities of ALP and GSH-Px were investigated. The results showed that the thymus and spleen index of SPD treated groups were significantly higher than those of PC, however, the num- ber of leucocytes and activity of ALP were lower than those of PC. There were no significant differences between Pre and Tre groups. SPD could inhibit the inflammation induced by LPS form the data displayed in the histopathological slide. Taken together, the above results indicated that SPD treatment can help to maintain oxidation/antioxidation balance and alleviate inflammation in rat mastitis induced by LPS.
