摘要
目的研究直肠癌相关基因的结构与功能。方法从人直肠癌cDNA消减杂交文库结合基因芯片筛选出一个差异表达的EST片断(基因登录号为ES274071),利用生物信息学方法对其进行结构与功能的分析。结果得到其全长cDNA序列并预测编码蛋白为SETprotein,它是蛋白质磷酸脂酶2A及肿瘤抑制因子NM23的抑制剂。结论SET可能参与DNA转录的激活和DNA损伤的修复,与直肠癌的发生发展可能存在有一定关系。
Objective To investigate the structure and function of genes related in human rectal cancer. Methods Filtering through suppression subtractive hybrization and eDNA chip technique, we selected one differential EST segment in rectal cancer from normal tissue (whose accession number is ES274071 ) ,and its structure and function were analyzed by the method of bioinformatics. Results The total eDNA sequence was obtained, and the protein coded by it was predicted as SET protein which is a suppressor of protein phosphatase 2A(PP2A) and non-metastatic gene( NM23 ). Conclusion SET may play a role in activation of DNA transcription and recovery of DNA injury and is related in development of rectal cancer.
