摘要
目的:通过观察癫痫模型大鼠脑内单胺类递质水平的变化,探讨其在癫痫发病机制中的作用。方法:腹腔注射戊四氮(PTZ)建立Wistar大鼠癫痫模型,应用荧光分光光度法测定大鼠额叶、海马中多巴胺(DA)、5-羟色胺(5-HT)和去甲肾上腺素(NE)的含量。结果:癫痫模型组额叶中5-HT、DA均高于正常对照组(P〈0.05或P〈0.01);NE含量低于对照组,两者之间的差异有显著性(P〈0.01);海马中:癫痫模型组DA与正常对照组比较无明显差异,5-HT、NE的含量均低于正常对照组(P〈0.05或P〈0.01)。结论:脑内单胺类递质参与了癫痫活动调控,单胺水平的改变将导致脑的兴奋性改变。作用的性质取决于所在脑区,具体机制有待深入的研究。
Objective :To study the concentrations of Monoamine in epileptic rats kindled by Pentylenetetrazol and the involvement of neurotransmitter systems as epileptogenesis modulators. Methods:PTZ (35mg/kg) intraperitoneal injection every day to establish chronic PTZ - kindling model. The concentrations of Monoamine ( dopamine ( DA ) , serotonin ( 5 - HT ) and norepinephrine (NE) ) of frontal cortex and hippocampus were determined by flurospectrophotometry. Results : For the hippocampus determinations, 5 - HT ,NE concentrations of experimental model group were significantly lower than control group ( P 〈 0.05 or P 〈 0.01 ) ; There were no significantly dereference of the concentrations of DA between two groups ( P 〉 0.05 ). whereas, For the frontal cortex determinations , 5 - HT,DA concentrations of model group elevated significantly as compared with control group (P 〈 0.05 ,P 〈 0.01 ). Compared with control group, the concentrations of NE in experimental model group decreased (P 〈 0.05 ). Conclusions: These results indicate that monoamine takes part in the epileptic process; And its effect on seizure depend on the brain area studied. Different mechanisms involved need to be further studied.
