Significant and Expression of Matrix Metalloproteinase-2 and Vascular Endothelial Growth Factor in Cutaneous Squamous Cell Carcinoma

摘要 目的探讨皮肤鳞状细胞癌(简称鳞癌)中基质金属蛋白酶2(matrix metalloproteinase 2,MMP-2)、血管内皮生长因子(vascular endothelial growth factor,VEGF)的表达及其与肿瘤分化和淋巴结转移的关系。方法用ABC免疫组化技术检测50例皮肤鳞癌标本(其中高分化鳞癌34例,低分化鳞癌16例,伴有淋巴结转移14例)中MMP-2、VEGF表达。结果MMP-2、VEGF主要表达在癌巢中心,表达阳性率分别为76%、72%;二者在低分化鳞癌中的表达明显高于高分化者(P<0.05),且其表达越高,淋巴结转移率越高(P<0.05)。结论皮肤鳞癌中MMP-2和VEGF表达与其分化程度及淋巴结转移有关。 Objective To investigate the expression of matrix metalloproteinase-2(MMP-2) and vascular endothelial growth factor(VEGF) in cutaneous squamous cell carcinoma(SCC) and the relationship between their expression and tumor differentiation and lymph node metastasis. Methods The expression of MMP-2 and VEGF was observed by ABC imunohistochemiccal techique in 50 cutaneous SCC(including 14 SCCs with lymph node metastases). Among them, there were 34 highly differentiated ones and 16 poorly differentiated ones. Results MMP-2 and VEGF were localized within cytoplasm of tumor cells. MMP-2 expressive rate was 76%, and expression in poorly differentiated tumors was significantly higher than that in highly differentiated ones(P=0.021), and the higher expression of MMP-2, the higher rate of lymph node metastasis(P= 0.003). VEGF expressive rate was 72%. The expression in poorly differentiated tumors was significantly higher than that in highly differentiated ones(P=0.0389), and the higher expression of VEGF, the higher rate of lymph node metastasis(P=-0.0168). Conclusion Expression of MMP-2 and VEGF is relate to the differentiation and lymph node metastasis in cutaneous SCC.
