摘要 目的观察PTD-BDNF在实验动物体内对脑缺血再灌注损伤的治疗效果。方法线栓法建立大鼠大脑中动脉闭塞模型。PTD-BDNF组于缺血再灌注后静脉注射PTD-BDNF2mg/kg,对照组注射等体积生理盐水。采用TTC染色、HE染色及TUNNEL染色分析PTD-BDNF对脑缺血再灌注损伤的神经保护作用。结果在大鼠局灶性脑缺血再灌注模型中,与对照组相比,PTD-BDNF融合蛋白使脑缺血梗死体积百分比从18.20%降至10.12%(n=4,P<0.01)。HE染色可见PTD-BDNF组脑组织缺血坏死程度明显减轻,细胞核皱缩、碎裂现象减少。TUNNEL染色显示梗死灶周边平均每个视野TUNNEL阳性细胞率,从51.24%降至23.54%(n=6,P<0.01)。结论PTD-BDNF融合蛋白经静脉给药后能通过血脑屏障发挥神经保护作用,减轻脑缺血再灌注损伤,改善神经功能。 Objective To observe the in vitro effects of PTD-BDNF on ischemia reperfusion injury in rats. Methods A rat model of middle cerebral artery occlusion (MCAO) was established by intraluminal filament occlusion with thread ligation. Rats in PTD-BDNF group were intravenously injected with 2mg/kg PTD-BDNF immediately after ischemia reperfusion, and rats in control group were injected with the same volume of normal saline. TTC, HE and TUNEL staining were used to analyze the neuroprotective effect of PTD-BDNF on ischemia reperfusion injury in rats. Results Compared with the control group, the percentage of infarct volume and TUNEL positive rate decreased from 18.20% to 10.12% and from 51.24% to 23.54% , respectively, in PTD-BDNF group ( n = 4, n = 6, P 〈 0.01 ), implying that PTD-BDNF can significantly decrease the infarction volume and apoptosis ratio, and increase the survival time of neurons in rats with focal cerebral injury induced by ischemia-reperfusion. Conclusion Intravenous PTD-BDNF can protect neurons from injury across the blood brain barrier, reduce ischemia reperfusion injury and improve the function of cranial nerves.
