摘要
目的研究速灭威对N2a成神经元细胞、C6胶质细胞和小鼠胚胎神经干细胞增殖的影响。方法用四噻氮唑盐比色法观察速灭威在不同时点(12、24、36h)对C6、N2a及小鼠胚胎神经干细胞增殖的影响。结果速灭威对C6胶质细胞、N2a成神经细胞和小鼠胚胎神经干细胞增殖均有明显抑制作用,且呈现剂量和时间依赖性;5mg/L的速灭威作用24h就可明显抑制小鼠胚胎神经干细胞增殖。3种细胞对速灭威的增殖抑制作用敏感性依次为mNSCs>C6>N2a。结论速灭威可以抑制C6胶质细胞、N2a成神经细胞和小鼠胚胎神经干细胞增殖,且小鼠胚胎神经干细胞对速灭威敏感性最强。
Objective To explore the effects of m-tolyl methylcarbamate (MTMC) on cell proliferation of mouse neuroblastoma (N2a) cells ,C6 glioma cells and mouse embryonic neural stem cells and compare their differences. Methods Dose- and time-dependent inhibitory effected of m-tolyl methylcarbamate on cell proliferation of mouse neuroblastoma(N2a) cells ,C6 glioma cells and mouse embryonic neural stem cells (mNSCs) were detected using MTF assay. Results MTMC inhibited the proliferation of mouse neuroblastoma (N2a) cells, C6 glioma cells and mouse embryonic neural stem cells remarkably in a dose-and time-dependent manner, and even 5 mg/L MTMC could inhibit the proliferation of mouse embryonic neural stem cells after 24 h treatment, and the order of the sensitivity of them to MTMC was mNSCs 〉 C6 〉 N2a. Conclusion MTMC can inhibit the proliferation of mouse neuroblastoma(N2a) cells, C6 glioma cells and mouse embryonic neural stem cells, and mouse embryonic neural stem cells are the most sensitive to MTMC.
