表皮生长因子对肝星状细胞c—jun基因表达的影响

摘要 目的观察表皮生长因子(EGF)对肝星状细胞c-jun基因表达的影响。方法在培养的肝星状细胞株中加入不同浓度的EGF(终质量浓度20、100、500μg/L),于8、24、48、72h四个时间点分别收集细胞,提取肝星状细胞总RNA;用逆转录定量聚合酶链反应(PCR)方法测定c-jun的基因表达水平。结果EGF3组HSC—T6细胞c-jun基因表达水平在8h(1.46±0.16、2.12±0.20、2.78±0.32)、24h(1.32±0.13、1.87±0.17、2.43±0.26)、48h(1.03±0.10、1.51±0.13、2.03±0.20)、72h(0.77±0.09、1.17±0.10、1.66±0.12)四个时间点均显著高于对照组;随剂量加大,HSC—T细胞c-jun基因表达水平逐渐升高;3组间差异有统计学意义。结论EGF对肝星状细胞c-jun基因表达有明显的上调作用。 Objective To investigate the effects of epidermal growth factor (EGF) on c-jun gene expression in hepatic stellate ceils. Methods Hepatic stellate cells (HSC-T6) were cultured in medium containing different concentrations of EGF ( 20,100, and 500 μg/L respectively), and collected at different incubation periods (8,24,48 ,and 72 h). Total RNA of HSCs was isolated,and c-jun gene expression levels were measured by reverse transcription polymerase chain reaction. Results C-jun gene expression levels of HSCs in low (20 μg/L) ,middle ( 100 μg/L) and high (500 μg/L) EGF groups were significantly higher than those of control group after exposure to EGF at 8 h ( 1.46 ± 0. 16,2. 12 ± 0.20,2.78 ± 0.32) ,or24 h (1.32 ±0.13,1.87 ±0.17,2.43 ±0.26) ,or48 h (1.03 ±0.10,1.51 ±0.13,2.03 ± 0.20 ), or 72 h (0.77 ± 0.09,1.17 ± 0.10,1.66 ± 0.12 ), respectively. The c-jun gene expression levels of HSCs were enhanced gradually with the increased dosage of EGF, and there were significant differences in the c-jun gene expression among three EGF groups. Conclusion EGF may strongly up-regulate c-jun gene expression in HSCs.
