大鼠角膜碱烧伤新生血管形成中β-catenin的表达

摘要 目的:探讨检测大鼠角膜碱烧伤后新生血管形成过程中β-连环蛋白(β-catenin)的表达变化。方法:使用碱烧伤诱导左眼角膜新生血管模型,SD大鼠25只随机分成5组,使用碱烧伤诱导左眼角膜新生血管模型,将右眼作为对照组,每日裂隙灯下观察角膜新生血管的发展,并采用免疫组织化学和逆转录-聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)方法,分别检测碱烧伤1,4,7,14,21d组以及正常对照组β-catenin和血管内皮生长因子(vascula rendothelial growth factor,VEGF)的表达。结果:在正常大鼠角膜组织中,β-catenin不表达或弱表达,碱烧伤后1d时,β-catenin免疫活性开始增强,4d时表达最强,14d时下降,21d时仍有弱表达。RT-PCR显示碱烧伤后β-catenin表达增强,在伤后4d表达最明显,21d后呈极弱表达。VEGF与β-catenin表达分布相似,强度较弱,二者之间呈正相关(r=0.855,P<0.05)。结论:角膜碱烧伤后,β-catenin表达水平与新生血管形成明显相关性。 AIM: To investigate the expression of β-catenin in cornea after alkali burn and explore its role in corneal neo- vascularization. METHODS: Corneal neovascularizalion (CNV) model was established by putting filter paper with the size of 3 mm in diameter immersed in lmol/L NaOH solution on the rat left cornea for 20 seconds. Twenty-five Sprague Dawley rats were randomly divided into 5 groups, post- operation 1 day, 4, 7, 14 and 21 days group while right eyes as normal control group. The expression level of β-catenin protein and mRNA was determined at 1 day, 4, 7, 14, 21 days following establishmend of model by RT- PCR and immunohistochemical technique. RESULTS: There was no evident β-catenin immunoreactivity in normal cornea. Both expression of β-catenin and vascular endothelial growth factor(VEGF) peaked in 4 days and gradually decreased to near baseline 21 days later. Al- teration of β-catenin and VEGF levels showed a significant positive correlation ( r = 0.855, P 〈 0.05). CONCLUSION: The levels of β-catenin were markedly related to inflammatory corneal neovascularization in rat cornea after alkali burn.
