摘要
目的:探讨天麻素对戊四氮(PTZ)致痫大鼠脑内血管的保护作用及可能机制。方法:102只Wistar大鼠,随机分成对照组(6只),PTZ致痫组(PTZ组,24只),丙戊酸钠干预组(VPA组,24只),小剂量天麻素干预组(Gs组,24只)和大剂量天麻素干预组(Gb组,24只),每组再分4个时间点(12h、48h、5d和7d)。运用免疫组化法检测大鼠海马区基质金属蛋白酶-9(MMP-9)、CD34的变化。结果:除对照组无痫样发作外,其余各组全部出现Racine分级Ⅳ~Ⅴ级痫样发作。PTZ组血管内皮细胞以及周边细胞均有明显的TUNEL染色阳性表达,而干预组阳性细胞显著减少。MMP-9染色在PTZ组海马区血管有高表达,与VPA组、Gs组和Gb组比较IOD差异有统计学意义(P<0.001)。PTZ组CD34表达水平增高明显,与VPA组、Gs组和Gb组比较MVD差异有统计学意义(P<0.001)。结论:大、小剂量天麻素和丙戊酸钠都能减轻PTZ致痫大鼠海马区血管的损伤,两种药物疗效相当。
Objective To explore the protective effects and possible mechanism of gastrodin on cerebral vascular of rats with seizures disorder induced by pentylenetetrazol (PTZ). Methods 102 Wistar rats were divided into control group (n = 6), PTZ group (n = 24), sodium valproate (VPA) group (n = 24), low dose gastrodin group (Gs group, n = 24), and high dose gastrodin group (Gb group, n = 24). Each group was further classified into 4 subgroups. The expressions of terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL), matrix metalloproteinase- 9 (MMP-9), and CD34 in hippocampus were measured by immunohistochemistry. Results Except the control group, seizures disorder (Racine Ⅳ-Ⅴ class) was shown in the other 4 groups. TUNEL was highly expressed in vascular endothelial cells and peripheral cells in PTZ group, and was lowly expressed in VPA, Gs, and Gb groups (P 〈 0.001 ). MMP-9 and CD34 were highly expressed in PTZ group, and the expressions were significantly different from those in VPA, Gs, and Gb groups (P 〈 0.001). Conclusion Gastrodin and VPA have the similar effect of reducing the damage of vascular in the hippocampus of rats with seizures disorder induced by PTZ.
