半夏泻心汤防治化疗药致胃肠反应机制的实验研究 被引量:8

摘要 目的:探讨半夏泻心汤防治化疗药顺铂(DDP)所致胃肠道反应的机制。方法:本实验用静脉注入DDP的方法制造实验性胃肠道反应大鼠模型,采用内置气囊法测定大鼠胃内压力;采用比色法测定大鼠十二指肠组织中SOD及MAO活性;采用荧光分光光度法测定大鼠脑组织中5-HT含量;应用透射电镜观察大鼠胃窦组织中嗜铬细胞病理。结果:①静脉注射DDP后,与正常对照组比较,模型组胃内压力显著升高(P<0.05),而中药组胃内压力无显著变化(P>0.05),②静脉注射DDP后,与正常对照组比较,模型组及中药组肠组织中SOD、MAO活性下降,但中药组SOD、MAO活性下降低于模型组(P<0.05);③静脉注射DDP后,与正常对照组比较,模型组及中药组肠组织中SOD、MAO活性下降,但中药组SOD、MAO活性下降低于模型组(P<0.05);④电镜结果显示:静脉注射DDP后,与模型组比较,中药组大鼠胃窦组织中嗜铬细胞中分泌颗粒数量多,且电子密度高,与正常对照组比较无显著差异。结论:半夏泻心汤通过提高大鼠肠组织中SOD、MAO活性,抑制胃肠组织中嗜铬细胞内分泌颗粒量减少;减少5-HT释放,抑制DDP导致的胃内压力增高。 Objective :To study the mechanisms of Pinelliae decoction for purging stomach -fire preventing gastrointestinal (GI) tract reaction caused by cisplatin administration in rats. Methods : Using pathological technique, electrophysiological technique and biochemical technique to compare EC cells, activity of SOD and MAO, the concentration of 5 - HT and IGP in three groups rats ( the control group, MS group, TCM group) before or after DDP administration. Results : (1)After DDP administration, as compared with the control group, IGP in NS group was marked higher( P 〈 0. 05 ) , but showed no obvious change in TCM group.(2)After DDP administration, comparing with the control group, activity of SOD and MAO was lower in TCM group and NS group,but the activity of SOD and MAO in TCM group was higher than that in NS group ( P 〈 0. 05 ). (3)Comparing with the control group, the concentration of 5 - HT in the brainstem were higher in TCM group and NS group,but lower in TCM group than that in NS group(P 〈0. 05). (4)After DDP administration,the quantitative and electron-dense of secretory granules of EC cells in TCM group were higher than that in NS group. There was no significant difference between the change of EC cells in TCM group and that in the control group. Conclusion : (1)IGP increased after DDP administration. (2)Activity of SOD and MAO in the duodenum decreased after DDP administration. (3)Concentration of 5 - HT in the brainstem increased after DDP administration. (4)Quantitative and electron - dense of secretory granules of EC cells in stomach decreased after DDP administration. The mechanisms of chemotherapy-induced GI tract reaction may be due to 5 - HT release. The mechanisms of Pinelliae decoction for Purging Stomach - fire preventing GI tract reaction induced by chemotherapy maybe that it can inhibit the release of 5 - HT by protecting the intestinal mucosa.
