摘要
[目的]研究热带土壤解磷微生物种群结构,筛选鉴定高效解磷菌株,为热带土壤解磷细菌的筛选和菌肥制备奠定基础。[方法]以有机磷液体培养基进行生物富集,无机磷固体培养基平板法从海南、广东、云南等地热带土壤中筛选出解磷效果较好的一株解磷细菌PSB13。[结果]通过细菌16 S rRNA通用引物PCR扩增该菌株的16 S rDNA序列,测得序列长度为1 088 bp,经B lastn搜索进行序列比对,结果表明,该菌株属于Burkholderiasp。进一步的室内平板对峙培养试验表明,PSB13对Fusarium oxysporum piperis等11个供试病原菌中的9个具有不同程度的拮抗作用。[结论]PSB13是一株高效的具有开发前景的热带土壤解磷菌株。
[Objective] The basis of the isolation of the bacteria solving phosphorus from tropical soil and the preparation of bio-bacteria fertilizer was laid through the research on the structure of microbiology and selection and identification of the bacteria strain with the ability in th phosphorus-solving, [ Method ] The organic phosphobacteria liquid medium was used for biological enriching culture. A bacteria strain-PSB13 with the high-efficiency ability in the phosphorus-solving was screened from the sample of tropical soils from Hainan, Guangdong and Yunna province with the method of inorganic plate solid culture. [ Result] The 16 S rDNA sequence of the strain was amplified with PCR techniqu, and its length was 1088bp. Compared with Blastn, the result showed that the strain belonged to Burkholderia sp. The results of in-door anta.g onism experiment indicated that the PSB13 had antagonism reaction to 9 of the 11 pathogens 11 strains such as Fusarium oxysporum piperis.[Conclusion] The PSB13 was one strain with he high-efficiency ability in the phosphorus-solving, which could have the good viewpoint of utilization and development.
