摘要
目的研究CDC6、HSP27在宫颈癌组织中的表达及其与HPV16/18感染的相关性及其临床意义,探讨宫颈癌的发生机制。方法采用免疫组织化学方法检测50例宫颈癌、20例宫颈上皮内瘤变(cervical intraepithelial neopla-sia,CIN)CINⅠ、20例CINⅡ-Ⅲ、20例正常宫颈组织石蜡样品中CDC6、HSP27的表达,同时采用PCR技术检测HPV16/18感染情况。结果在宫颈癌组织中CDC6、HSP27的阳性率高于CIN和正常宫颈组织(均P<0.05),且CDC6、HSP27阳性表达率与病理分级和淋巴结转移有关(均P<0.05);HSP27阳性表达率与临床分期有关(P<0.05),CDC6阳性表达率与临床分期无关(P>0.05);在宫颈癌组织中CDC6和HSP27的阳性表达率与年龄分组无关(均P>0.05)。②HPV16/18在正常宫颈组织、CIN和宫颈癌中的阳性率逐渐升高(P<0.05),但与临床分期、病理分级、淋巴转移、年龄分组无关(均P>0.05)。③HSP27和CDC6、HPV16/18的表达无相关性(均P>0.05);CDC6与HPV16/18感染有关(P<0.05)。结论CIN及宫颈癌中CDC6表达的改变可能与HPV16/18感染有关,共同影响CIN的发展及宫颈癌的发生和发展。
Objective To investigate the relationship between HPV16/18 infection and expressions of CDC6 as well as HSP27 in uterine cervical carcinomas. Methods Formalin - fixed, paraffin - embedded samples including 50 eases of cervical squamous eell carcinoma, 20 cases of cervical intraepithelial neoplasia (CIN) grade Ⅱ/Ⅲ and 20 eases of CIN grade Ⅰ and 20 eases of normal cervical tissues were studied. Using immunohistochemieal technique, the expressions of CDC6 and HSP27 in these samples were examined. The infections of HPV type 16, 18 DNA were determined by PCR Results ①There were significant differences in CDC6 and HSP27 expressions between carcinoma and CIN, as well as normal cervices ( all P 〈 0. 05). Expressions of CDC6 and HSP27 were correlated with tumor grades and lymph node metastasis ( all P 〈 0. 05 ), and weren't correlated with age ( all P 〉 0. 05). Expression of HSP27 was correlated with clinical stages ( P 〈 0. 05 ), but that of CDC6 was not ( P 〉 0. 05 ). ②The positive rate of HPV type 16/18 was gradually increased in the order of normal tissues, CIN and carcinoma (P 〈 0. 05 ), and was not associated with age, tumor grades, clinical stages and lymph node metastasis (all P 〉 0. 05).③HSP27 was not correlated with the expression of CDC6 and the positive rate of HPV16/18 (all P 〉 0. 05 ). The positive rate of HPV16/18 was positively correlated with the expression of CDC6 ( P 〈 0. 05 ). Condusion The results suggest that changes of CDC6 expression in tissues of CIN and cervical carcinoma may be associated with HPV16/18 infection, which can influence the progression of CIN and cervical carcinoma.
