大孔吸附树脂纯化桑黄黄酮的研究 被引量:3

Study on Purification of Flavonoids from Phellinus Baumii with Macroporous Resin

摘要 研究4种大孔吸附树脂对桑黄黄酮类物质的吸附和解吸特性,筛选出吸附解吸率较高且耗时较短的树脂DM-301,并对其静态和动态的吸附解吸特性做了研究。优化出的最佳工艺参数为上样流速2 mL/min;在上样浓度为0.87 mg/mL的条件下,上样体积为7.5 BV(柱床体积);洗脱剂80%乙醇;洗脱速度5 mL/min;洗脱体积3倍柱床体积,上样液pH为自然pH(3.65);柱温为室温。 The adsorption and desorption rates of flavonoids from Phellinus baumii by four kinds of macroporous resins were investigated. It was found that DM-301 resin exhibited higher adsorption and desorption capability and took less time. So it was suitable for enrichment of flavonoids. The effect of pH, temperature and flow rate on the adsorption and desorption rates of DM-301 resin in static and dynamic experiment were also studied. The optimal parameters for the resin was that adsorption flow rate was 2 mL/min, adsorption volume was 7.5 BV( extract solution at a concentration of 0. 87 mg/mL), the desorption solvent were 80% ethanol, the eluent rate was 5 mL/ min, the eluent volume was three times bed volume, the pH of sample was natural pH (3.65) and the experiment was done at ambient temperature.
