摘要
运用SDS-聚丙烯酰胺凝胶电泳技术,结合Schiff试剂的糖蛋白染色法,检测了12种经济贝类(皱纹盘鲍、杂色鲍、嫁虫戚、泥螺、泥蚶、紫贻贝、虾夷扇贝、海湾扇贝、青蛤、文蛤、硬壳蛤、缢蛏)性腺组织中糖蛋白的表达特征。结果表明,所检测的贝类两性性腺中糖蛋白电泳主带迁移率很相近,其中各种贝类精巢多数表现为迁移率大致相同的2个条带,而卵巢中则表现为迁移率相近的2或3条主带,并且亲缘关系越近的贝类之间的电泳谱图相似性越高,该结果很好地说明了各贝类之间存在着或远或近的同源性;此外,不同贝类性腺的糖蛋白图谱仍旧存在着很大的差别,这充分体现了海洋经济贝类的生物多样性。
Glycoprotein, a kind of important physiological active substance, plays a necessary role in recognition of animal gametes. So glycoprotein expression was analyzed in gonads of 12 economic shellfishes (abalones Haliotis discus hannai , Haliotis diversicolor , Cellana toreuma , Bullacta exarata , Tegillarca granosa , mussle Mytilus edulis , yesso scallop Patinopecten yesoensis ,scallop Argopecten irradians , Cyclina sinensis , Meretrix rneretrix , Mercenaria rnercenaria , and Sinonovacula constricta ) by SDS-polyacrylamide gel and staining with Schiff reagent. The results showed that the mobility of glycoprotein electrophoretic main band in gonads of 12 economic shellfishes showed the similar pattern. There were 2 main bands with similar mobility in the spermary, but 2 or 3 in the ovary. The similarity of electropherogram seemed high in the closed genetic relationship, indicating that there was a definite homology between these shellfishes. However, the diversity in different shellfish electropherograms indicated the biodiversity of marine economic shellfishes.
