加味五子衍宗方有效部位对β-淀粉样肽致大鼠行为学改变的影响 被引量:12

Effect of Modified Wuzi Yanzong Prescription and its Components on Spatial Behavior in Rats Induced by β-amyloid Peptide 25-35

摘要 目的:观察加味五子衍宗颗粒及其有效组分对β-淀粉样肽所致老年性痴呆(AD)模型鼠空间学习记忆能力的影响。方法:采用大鼠侧脑室立体定向注射Aβ25-35的方法建立AD动物模型,服用加味五子衍宗颗粒及其总黄酮、总多糖15d后以全自动Morris水迷宫实验检测大鼠定位航行试验及空间探索试验,来评价大鼠空间学习记忆功能。结果:加味五子衍宗颗粒及其有效组分总黄酮、总多糖均能明显缩短模型大鼠在Morris水迷宫定位航行试验中的逃避潜伏期和搜索距离(P<0.05),显著增加空间探索试验中原平台所在象限搜索时间和距离占总时间和总距离的百分比(P<0.05,P<0.01)。结论:加味五子衍宗颗粒及其总黄酮、总多糖能改善由β-淀粉样肽所致AD模型鼠的空间学习记忆能力。 Objective: To observe the effect of Modified Wuzi Yanzong Prescription and its components on spatial behavior in the rats with Alzheimer' s disease (AD) by Aβ25-35. Methods: The animal model of AD was established by Aβ25-35 stereotactic injection into the lateral cerebral ventricle of rats. Rats were treated with Modified Wuzi Yanzong Prescription and its components (total flavonoids and total polysaccharodes) for 15 days, Morris water maze with the video record and computer system and the spacial exploration experiment were used to assess the behavior performances of the rats. Results: Significant differences were observed between model group and Modified Wuzi Yanzong Prescription and its components treated groups ( P 〈 0.05). Compared with the model group, the escape latency and the searching distance were markedly reduced ( P 〈 0.05 ), the percentage of the rats swimming time and distance in platform quadrant of the spacial experiment were markedly higher( P 〈 0.05, P 〈 0.01 )in Modified Wuzi Yanzong Prescription and its components treated groups. Conclusion: Modified Wuzi Yanzong Prescription and its component can improve learning and memory disorders of the model rats induced by Aβ25-35 .
