摘要
目的观察马钱子碱对裸鼠乳腺癌骨转移的抑制作用。方法复制乳腺癌MDA-MB-231骨转移裸鼠模型,20只荷瘤裸鼠随机分为4组:马钱子碱高剂量组(A)、中剂量组(B)、低剂量组(C)和对照组(D),分别予6.90、3.45、1.73 mg.kg-1和生理盐水,腹腔注射。观察各组裸鼠生存状态和肿瘤生长情况,绘制肿瘤生长曲线,评价抑瘤率;病理切片观察肿瘤细胞形态和骨转移情况。结果马钱子碱处理组(A、B、C)与对照组(D)相比,裸鼠生存状态良好,肿瘤生长受到抑制,各组的抑瘤率分别为A组61.4%、B组50.5%、C组17.5%(P<0.05)。结论马钱子碱对乳腺癌骨转移的生长有抑制作用,值得进一步研究其作用机制。
Aim To investigate the inhibiting effects of brucine on a bone metastasis model of breast cancer. Methods A bone metasta- sis model of breast cancer was established by injection of breast cancer cell line MDA-MB-231 into the right leg of nude mice proximally to the femur, then 20 mice were randomly divided into four groups, blank control group and brucine at high/middle/low dose (6.90,3. 45,1.73 mg · kg ^- 1ip ) group. Then growth inhibition rate and histological assay were applied to evaluate effects of brucine. Results Brucine groups resulted in a significant inhibition on the growth of the tumor and bone injury,with growth inhibition rate of 61.4% ,50. 5% and 17.5% respectively(P 〈 0.05). Conclusion Brucine can inhibit tumor growth,and alleviate bone injury.
