摘要
目的拟发现中国羊种布鲁氏菌omp2基因PstⅠ酶切多态性特征。方法用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)对羊种标准菌株、疫苗株M5、国内分离的羊种布鲁氏菌野毒株及非典型株共46株进行分析。结果omp2基因扩增产物经PstⅠ酶切后呈现两种酶切图谱,43/46有238 bp和44 bp 2个条带;仅羊1型标准株16M和两株青海非典型羊种1型菌有282 bp、238 bp和44 bp 3个条带。结论omp2基因PstⅠ酶切可为区分部分野生动物中的羊种菌和家畜羊种菌,为揭示布鲁氏菌的遗传衍化关系提供线索。
Objective To explore PstⅠ digestion polymorphism of omp2 gene of Brucella melitensis isolated in China.MethodsStandard strain of Brucella melitensis,vaccine strain(M5),wild strain of Brucella melitensis isolated in China and atypical strain were analyzed by polymerase chain reaction-restriction fragment length polymorphism analysis(PCR-RFLP).ResultsThe amplification product of omp2 gene had two PstⅠ digestion pattern.Of the 46 strains analyzed,43 had 2 straps of 238 bp and 44 bp,only 1 standard strain of Brucella melitensis biotype 1(16M) and 2 atypical strains of Brucella melitensis biotype 1 isolated in Qinghai had 3 straps of 282 bp、238 bp and 44 bp.ConclusionPstⅠ digestion of omp2 gene can distinguish some Brucella melitensis in wild animal and Brucella melitensis in cattle,which provide the evidence to reveal the geno-derivation of Brucella.
