亲骨转移乳腺癌细胞MDA-MB-231BO成瘤性的初步研究

摘要 目的通过比较亲骨转移乳腺癌细胞(MDA-MB-231BO)和亲代乳腺癌细胞(MDA-MB-231)的生长曲线和致瘤性,初步探讨MDA-MB-231BO细胞的生物学特性。方法MTT法测定两种细胞的生长曲线,并将两种乳腺癌细胞接种于裸鼠腋窝处皮下,建立乳腺癌细胞异种移植瘤动物模型,30 d后处死裸鼠,肿瘤组织及相关脏器官做病理检查。结果MTT法测得MDA-MB-231BO细胞生长速率高于MDA-MB-231细胞。接种两种乳腺癌细胞的裸鼠均长出肿瘤,成瘤率为100%。病理检查符合人乳腺癌细胞特征,MDA-MB-231BO组瘤体体积明显大于MDA-MB-231组(P<0.05)。结论MDA-MB-231BO细胞生长速率高于MDA-MB-231细胞,而且MDA-MB-231BO在裸鼠体内的致瘤性强于MDA-MB-231。 Objective To compare the growth curve and oneogenieity of bone-seeking human breast cancer cell strain (MDA-MB-231BO) and parental human breast cancer cell strain (MDA-MB-231), and then study biological characteristics of breast cancer cell MDA-MB-231BO. Method Growth curve of MDA-MB-231BO and MDA-MB-231 were observed by MTT method, and these two cells were inoculated to the nude mice to establish the transplanted model. After 30 days the nude mice were killed and the tumors and associated organs were detected by pathological examination. Result The growth rate of MDA- MB-231BO was higher than MDA-MB-231. The implanted tumors grow well and the rate of success was 100% . The result of pathology detection met with the characteristics of breast cancer, and the growth of the tumor was significantly reduced in size in mice injected with MDA-MB-231 compared with the MDA-MB-231BO(P 〈 0.05). Conclusion The growth rate of MDA-MB- 231BO was higher than MDA-MB-231, and oneogenieity of MDA-MB-231BO was obviously lower than MDA-MB-231.
