摘要
采用不同剂量的^(60)Co-γ辐照对琥珀酸产生菌MS-23的原生质体进行诱变处理,结果表明,辐照剂量为30kGy时致死率超过90%,具有很好的诱变效果。经多次筛选得到1株高产琥珀酸的突变株MSCo-24,高效液相色谱法测定其琥珀酸产量为2.08g/L,比出发菌株产酸量提高了63.78%,遗传性状稳定。
Different dosages of ^60Co-γ ray were used to treat the protoplasts from spores of the succinic acid-producing strain MS-23. The results showed that when mutagenesis dosage of 30,000 Roentgen was used, the fatality rate reached above 90% , and a good mutagenic effect occured. By re-screening, a high-yielding mutant strain MSCo-24 was obtained. Its production of succinic acid, which was measured by high performance liquid chromatography (HPLC) , reached 2.08g/L, about 63.78% higher than that of the parent strain, and the genetic character was stable.
