葡萄糖基-β-环糊精作为手性选择剂对苯磺酸氨氯地平的毛细管电泳手性拆分

Enantioseparation of Amlodipine Besylate Using Glucosyl-β-cyclodextrin as Chiral Selector by Capillary Electrophoresis

摘要 以葡萄糖基-β-环糊精(Glu-β-CD)为手性选择剂,用毛细管区带电泳法对手性药物苯磺酸氨氯地平进行了拆分研究。考察了缓冲液的pH值、缓冲液浓度、缓冲液体系组成、Glu-β-CD的浓度及电压等对分离的影响,并对3批市售左旋苯磺酸氨氯地平片(施慧达)进行光学纯度检查。结果表明,在背景电解质为含20mmol/LGlu-β-CD的200mmol/L乙酸-三乙醇胺(HAc-TEA)(pH4.0)体系,电压25kV,温度20℃,检测波长214nm的条件下,苯磺酸氨氯地平可以得到良好分离,分离度为4.0。 A capillary zone electrophoresis method was developed for the separation of amlodipine besylate enantiomers using glucosyl-β-cyclodextrin(Glu-β-CD) as the chiral selector. Effects of pH val- ues, concentrations and compositions of buffer, concentrations of Glu-β-CD, and the applied voltage on the enantiomeric separation were investigated. The optimized conditions were using pH 4. 0 200 mmol/L acetic acid -triethanolamine solution as background electrolyte, 20 mmol/L Glu-β-CD as chiral selector, applied voltage of 25 kV, capillary temperature of 20℃ and detection wavelength of 214 nm. Under the optimal enantioseparation conditions, enantiomers of amlodipine besylate were completely separated with a resolution factor of 4.0 in 10 min. The method was rapid and simple, and was applied in the optical purity analysis of S-( - )- amlodipine besylate on market (Shihuida) with satisfactory results.
