摘要
以氨基糖苷类抗生素阿米卡星作为手性选择剂,在普通熔融石英毛细管上对后马托品进行了毛细管手性拆分研究。考察了影响分离的因素:诸如阿米卡星的浓度、背景电解质的种类和pH、有机添加剂的种类和含量、分离电压和毛细管柱温度等。通过优化实验条件,在30mmol/L的磷酸盐(含15mmol/L阿米卡星,30%(φ)甲醇)缓冲溶液中,分离电压为14kV,温度24℃,使后马托品对映体得到了拆分。结果表明阿米卡星可以作为一种新的手性选择剂应用于毛细管电泳手性拆分。
The enantiomers of homatropine were separated by capillary electrophoresis on a fused silica capilˉlary using aminoglycoside antibiotics amikacin as a chiral selector.The effects of the amikacin concentration,type and pH of background electrolyte,type and content of organic modifier,separation voltage,and capillary columntemperature onthe enantiomeric separationwere investigated.Optimumseparation conditions forhomatˉropine enantiomers were obtained as follows:amikacin,15mmol/L;concentration of NaH 2 PO 4 ,30mmol/L;methanol content,30%(φ);separation voltage,14kV;pH,9.0;and temperature,24℃.The results indiˉcated that amikacin was a newefficient chiral selector for capillary electrophoresis enantioseparation.
