摘要
目的:观察红背丝绸2种提取物和单体(CA1201)对内皮素1(endothelin1,ET1)生物效应的拮抗作用。方法:用红背丝绸3种提取物对ET1致小鼠急性死亡、血管收缩和升压作用的拮抗,探索用中草药拮抗ET的生物效应。结果:3种提取物对ET1致小鼠急性死亡有明显的保护作用,使小鼠的死亡率下降;并可拮抗ET1致胸主动脉的缩血管效应,醇提物和乙酸乙酯提取物(10-7母液)拮抗ET缩血管效应分别为57.27%和58.61%;CA120110-8mol·L-1和10-9mol·L-1亦能拮抗ET1的缩血管效应;醇提物和乙酸乙酯提取物ig及CA1201iv后能拮抗ET1的升压作用,拮抗ET1作用均呈现明显的剂量依赖性。
OBJECTIVE: To observe the antagonism effects of both alcohol and ethyl acetate extracts, monomer CA 1201 three components from Chinese herb Cissus assamica on endothelin 1 (ET 1) biological responses in vivo and in vitro. METHODS: We used both extracts and monomer CA 1201 to antagonize mouse acute death and vasoconstriction and blood pressure elevation induced by ET 1. RESULTS: These components could protect the acute death induced by ET 1 with reduce of death rate; and antagonize the vasoconstriction induced by ET 1 on isolated arotic ring of rats; both alcohol and ethyl acetate extracts through oral administration, CA 1201 iv route could blunt effects of elevation of blood pressure induced by ET 1, the effects of these components were in dose dependent manner. CONCLUSION: Both extracts and monomer CA 1201 from Chinese herb Cissus assamica can antagonize ET 1 biological effects.
