摘要
目的探讨阿魏酸钠(SF)对糖尿病大鼠肾脏蛋白激酶C(PKC)活性的影响。方法链脲佐菌素诱导的糖尿病大鼠灌胃给予SF110mg·kg-1·d-1,治疗8周,测定各组大鼠肾皮质超氧化物歧化酶、过氧化氢酶、PKC活性及丙二醛含量,观察肾脏病理改变。结果与正常对照组相比,糖尿病对照组大鼠肾重/体重、肌酐清除率、24h尿蛋白、肾皮质胞膜PKC活性、胞膜PKC活性占总活性的百分比、肾皮质丙二醛显著升高,肾皮质超氧化物歧化酶、过氧化氢酶活性显著降低,肾脏病理学改变明显,SF治疗组上述指标显著改善。结论SF可通过抑制肾脏PKC的激活对糖尿病大鼠肾脏产生保护作用。
Objective To investigate the effects of sodium ferulate (SF) on the activity of renal protein kinase C (PKC) in diabetic rats. Methods The diabetic rats induced by streptozotocin were treated with SF (110 mg/kg) per day for 8 weeks. The level of malondialdehyde and the activity of protein kinase C (PKC), superoxide dismutase and catalase in renal cortex were measured. The pathological changes were also observed. Results The creatinine clearance rate, renal weight/body weight, proteinuria, the activity of membrane PKC in renal cortex and the rate of membrane PKC activity in total PKC activity in diabetic control group were significantly higher than that in normal control group. In the diabetic control group, there was a highter level of malondialdehyde and a lower activity of superoxide dismutase and catalase in renal cortex than that in normal control group. Compared with normal control group, the pathology changes were also significant in the diabetic control group. These abnormalities were significantly ameliorated in the diabetic rats treated with SF. Conclusion SF ameliorates diabetic nephropathy and inhibits the activation of PKC in kidney of diabetic rats.
