RhG-CSF对快速老化模型小鼠P10脑组织炎症反应的干预作用 被引量:1

RhG-CSF inhibits inflammatory reaction of brain tissues in senescence accelerated mouse P10

摘要 目的观察重组人粒细胞集落刺激因子(rhG-CSF)对快速老化模型小鼠P10(SAMP10)炎症反应的抑制作用。方法随机将48只SAMP10分为对照组、rhG-CSF治疗组,每组24只。治疗组皮下注射rhG-CSF100μg/(kg·d),对照组给予等体积生理盐水,连续7d。分别于给药结束后第7、14、28、56天取脑组织进行免疫荧光染色,检测TNF-α和iNOS的表达,用Image-proPlus图像分析软件,分析TNF-α和iNOS阳性细胞百分率、总面积及灰度值。结果治疗组小鼠脑组织额叶皮质TNF-α和iNOS阳性细胞百分率、总面积及灰度值明显减少(P<0.05)。结论rhG-CSF能明显抑制SAMP10脑组织炎症反应。 Objective To observe the effects of rhG-CSF on the inhibition of the inflammatory response of senescence accelerated mouse P10 (SAMP10). Methods Forty-eight SAMP10 were randomly divided into the control group and the rhG-CSF group. Mice in the rhG-CSF group were subcutaneously subjected to 100μg/(kg·d) rhG-CSF for 7 days, while mice in the control group were subjected to normal saline. Expressions of TNF-α and iNOS were determined by immunofluorescence on the 7th, 14th, 28th, and 56th day after administration. The percentage, area and density of the TNF-α positive cells and iNOS positive cells were analyzed by Image-pro Plus software. Results Compared with the control group, the percentage, area and density of the TNF-α positive cells and iNOS positive cells significantly declined in the rhG-CSF group. Conclusionc RhG-CSF can significantly inhibit the inflammatory response of SAMP10.
