摘要
目的研究UC大鼠的免疫功能的变化,包括细胞因子及T细胞亚群的变化。方法以TNBS/乙醇方法制备UC大鼠模型,检测UC大鼠细胞因子TNF-α、IL-2、IL-4及T细胞亚群的变化。结果模型组血清TNF-α、IL-2含量及外周血CD3+、CD4+细胞百分比在1、3、7周3个时间段均升高(P<0.05)。而血清IL-4含量在1、7周时UC大鼠与对照组比较差异无统计学意义(P>0.05),3周时出现降低,与其它组比较差异有统计学意义(P<0.05);CD8+细胞百分比较正常组降低(P<0.05);CD4+/CD8+比值明显高于正常组(P<0.05)。结论T细胞亚群及细胞因子的变化,说明UC大鼠存在的细胞免疫功能亢进,进而引发肠粘膜局部的变态反应。
Objective To investigate the variation of immune function in UC rats, including T cell subset, the cytoldne of TNF-α, IL-2,IL-4.Methods To bulid UC animal model by TNBS/alcohol,ELISA was used to clctcct the cytokine of TNF-α,IL-2,IL-4.Flow cytometry was used to detect the T cell subset. Results The content of TNF-α,IL-2 in blood serum was predominantly increased at the first, third and seven week in model group than in normal group ( P 〈 0.05 ). The content of IL-4 in blood serum hadn' t significant deviation at the first,seven week between model group and normal group ( P 〉 0.05) .The percentage of CD3^+ ,C, D4^+ in pefiphefial blood was predominantly increased at the first,third and seven week in model group than in normal group( P 〈 0.05) ,but the percentage of CIX+ was lower than normal group( P 〈 0.05 ), the ratio of CD4^+/CD8^+ was higher than normal group ( P 〈 0.05 ). Conclusion The results show the abnormal increasing of cell immunoresponse,it may cause the intestinal mucous activate the local allergy.
