摘要
目的使人工血管体外形成内皮衬里并评价其抗血小板沉积特性。方法将培养扩增的成人大隐静脉内皮细胞(AHSVEC)铺植于贫血小板血浆和纤维连接素预衬的聚四氟乙烯人工血管,并体外培养9天,再用含^(125)Ⅰ标记血小板的全血以100ml/min的流速灌注60分钟。人工血管内壁形态以扫描电镜观察,血小板沉积量以r计数测定。结果衬里的人工血管内壁形成了完全融合的内皮单层;血小板沉积量显著低于未衬内皮的对照组人工血管。结论内皮衬里的人工血管具有良好的抗血小板沉积特性。
Objective To assess the capacity of lined endothelial cell (EC)monolavers on small-caliber vascular grafts to prevent early platelet depsoition in vitro. Methods Cultured adult human saphenous vein ECs were sodded to polytetrafiuoroethylene grafts precoated with platelet-poor-plasma and human fibronectin, cultured for 9 days, and then perfused in vitro for 60 min at 100ml/min with human blood containing^(125)Ⅰ-labeled platelets. Morphology of EC lining was examined by SEM. Numbers of deposited platelets were calculated by gamma counting. Results EC lining on the vascular grafts completely formed confluent EC monolayers. Platelet deposition on the EC lined grafts was significantly reduced as compared with that on the untreated control grafts. Conclusion This approach to lining graft in vitro with adult human saphenous vein ECs may be helpful in decreasing early thrombus formation on smallcaliber vascular grafts.
